A DNA fragment encoding cre/ERT2 (cre recombinase fused to the human estrogen receptor with a mutated ligand-binding domain) was inserted downstream of a 6.3-kb fragment of the human CD19 promoter. This construct was inserted into the LC-1 BAC via Red-mediated homologous recombination in E. coli. The recombinant BAC was injected into the pronuclei of fertilized C57BL/6 oocytes. Two founders were generated and one line designated LC-1-hCD19-CreERT2 provided functional cre-mediated recombination. PCR analysis showed that animals carried a single copy of the transgene.
