Trapped gene association determined by genome coordinate overlap and strand. The gene trap vector inserted into intron 2. The genetrap allele initiates transcription from the endogenous promoter and prematurely terminates at the polyadenylation sequence in the FlipROSABetaGeo vector, resulting in a truncated mRNA that encodes a non-functional protein. RT-PCR of E11.5 embryos using primers flanking the gene trap insertion indicates very weak mRNA expression. However, RT-PCR using primers further towards the 3'-end (exons 3-7) indicates alternate transcripts. Quantitative real-time PCR that amplifies exons 4-5 shows that alternate transcripts are present at very low levels. Western blot analysis of sciatic nerves and mouse embryonic fibroblasts, however, shows no protein expression or expression of any smaller protein isoforms, indicating a null allele. (J:211808)
