A tissue-specific transgene encoding an inducible MYC protein was generated by coupling the mouse villin promoter (including 3.5 kb upstream of the transcription start site and 5.5 kb of the first intron) to a fusion cDNA encoding the human MYC protein joined to a modified mouse estrogen receptor (ER) ligand-binding domain. The Gly525Arg mutant ER domain (ERTM) responds to 4-hydroxytamoxifen (4OHT) but is insensitive to the endogenous ligand, 17beta-estradiol; therefore, MYC is active only in the presence of 4OHT. Immunofluorescence microscopic analysis confirmed the expression of the fusion protein throughout the intestinal epithelium of transgenic mice. (J:53387, J:118587, J:153653)
