Transgenic mice express prion protein in the cytosol (cyPrnP, amino acids 23 to 230, which precisely eliminated the NH2-terminal and the COOH-terminal sequences that are cleaved upon ER entry), and beta-Gal under the regulation of a minimal CMV promoter and a tetracycline-responsive bi-directional promoter. The whole brain beta-Gal staining and Western blot revealed that beta-Gal and cyPrnp expression were largely confined to the forebrain. (J:80582, J:153494)
