A cre recombinase sequence was targeted to the Trp73 locus in frame at the alternative start codon located in exon 3' (where transcription of the DeltaNp73 isoform is initiated), inactivating the DeltaNp73 isoform and leaving the primary Trp73 transcript (TAp73) unaffected. The cre sequence is followed by an internal ribosomal entry site (IRES) and an EGFP coding sequence. The targeting construct that was used also contains an FRT-flanked Neo cassette and a diphtheria toxin A (DTA) negative selection cassette. (J:153209)
