A floxed neo cassette was inserted upstream of exon 2 and a loxP site was inserted downstream of exon 3. However, further sequencing analysis revealed that the insertion of the loxp cassette disrupted the first splice site and resulted in an allele that, in contrast to the wildtype allele, did not result in expression of encoded protein.
