A 1.3-kb genomic segment containing both microRNAs Mir145 and Mir143 was replaced by a construct containing a lacZ reporter and a neomycin resistance cassette. Inactivation of the loci was confirmed by Northern blot analysis of homozygote aorta, bladder, liver and kidney. Reporter gene expression was detected in early embryonic heart development of heterozygotes, and in most smooth muscle cells of the adult heterozygote. (J:152702)
