A construct containing GFP and a floxed neomycin cassette were used to replace exon 1 of the gene locus. The neomycin cassette was removed after targeting. GFP expression was detected in the double-positive and single-positive thymocytes as determined by flow cytometry. Endogenous protein was not expressed in homozygote thymocytes. (J:152601)
