A floxed neomycin resistance cassette in reverse orientation was placed at the 3' end of intron 3. An additional loxP site was inserted just downstream of exon 4. In the resulting ES cells, exon 4 is flanked by loxP sites. These mutant ES cells were used in the generation of Sirt1tm1Fwa mice. (J:85542)
