A local hopping enhancer detector (LHED) construct was inserted into the 5' flank of the gene, over 49kb upstream of the translation start site. The LHED construct consisted of an frt site, a floxed Pgk1-neo cassette, a puromycin resistance gene, the sleeping beauty transposon pT2, a Pgk1 promoter, and a final frt site. The sleeping beauty transposon pT2 had been modified to contain a loxP site and the mouse Hspa1b minimal promoter fused to a lacZ gene. Removal of the transposon sequence results in expression of the puromycin resistance gene. Local hopping of the transposon allows for the creation of a nested series of deletion/inversion mutations via cre mediated recombination. (J:151776)
