Exons 1 and 2, which encode the transcriptional start site and the initiation ATG codon, were replaced by a PgkHPRT expression cassette. Correct targeting in HPRT-null ES cells was confirmed by Southern blotting. Gene inactivation was confirmed by immunoblot analysis on homozygous testes extracts. (J:152071)
