Mice carrying the Rpl22tm1.1Psam allele were crossed with Mox2-cre mice to remove the endogenous exon 4 from the germline. This allowed transcription of the knocked-in exon 4 that contains sequence encoding three hemagglutinin eptiopes upstream of the stop codon. Immunoblot analysis confirmed in homozygote tissue extracts that the mutant protein was expressed at levels similar to endogenous protein in controls. Mutant protein expression effectively "tags" ribosomal complexes with the HA epitope allowing for their isolation. (J:151962)
