An EYFP expression cassette and a floxed neomycin resistance cassette were inserted in-frame downstream of the last coding exon but upstream of the 3' untranslated region. Founder mice were crossed with cre-deleter mice to remove the neomycin cassette, leaving behind a loxP site. Immunoblot analysis of homozygote thymocyte extracts detected expression of just the fusion product. The fusion product was also detectable by flow cytometry. Developmental defects in a subset of homozygote NK T cells suggest some function of the endogenous protein is lost. (J:151699)
