A loxP site was inserted into intron 1 and an FRT site flanked neomycin resistance gene cassette and loxP site were inserted into intron 2. Exon 2, used for the TA isoforms, and the neo cassette were deleted through cre-mediated recombination. The absence of protein expression was confirmed by western blot analysis on keratinocyte extracts. This allele specifically knocks out the TA (trans activation domain) isoforms and leaves the deltaN isoform intact. (J:151638)
