The Hoxb7 promoter fragment from -1316 to +8 bp from the translation initiation codon into the CORE expression vector containing a cDNA encoding the reverse tetracycline-responsive transactivator (rtTA) sequence upstream of cDNA encoding cre recombinase controlled by a CMV minimal promoter linked to the tet operator (tetO) sequence. The construct was injected into the male pronucleus of fertilized oocytes. Eight founder animals were obtained and cre activity was analyzed by crossing the animals to R26R reporter mice. In this line, cre expression was restricted to cells at the midbrain-hindbrain boundary. The pattern of cre activity was not affected by doxycycline, indicating cre expression is not controlled the rtTA protein expressed from the Hoxb7 promoter. (J:135242)
