A gene trap vector (pGT0Lxf) containing a splice-acceptor sequence upstream of a beta-geo reporter gene and a polyA tail was inserted into intron 2 between nucleotides 64,308,443 and 64,316,218, resulting in deletion of the corresponding part of the gene including exon 3 (nt 64,312,349-64,312,475). Semi-quantitative RT-PCR and Western blot analysis confirmed the absence of mRNA and protein expression in E11.5 homozygous mutant embryos, while X-Gal staining revealed that expression of lacZ is detectable in E10.5 heterozygous and homozygous mutant embryos. All 31 exons 3' to the insertion site of the vector are not expressed. (J:277387)
