A gene-trap cassette containing a splice acceptor and a poly(A) signal was inserted between exons 2 and 3, so that a spliced transcript containing exons 1 and 2 (98 out of 1262 aa) is expressed instead of the full-length transcript. RT-PCR of MEFs showed reduced mRNA expression. Western blotting of spleen, liver, heart and kidney from homozygous mice failed to detect any wild-type protein. (J:157195)
