A gene trap vector randomly integrated in the intron between exons 18 and 19 of the gene. The gene trap cassette contains a splice acceptor that co-opts the endogenous splicing of the mRNA transcript, leading to an in-frame fusion protein containing the first 823 residues of the encoded protein and beta-galactosidase. This gene trap vector also provides a human placental alkaline phosphatase (PLAP) reporter gene expressed through an internal ribosome entry site (IRES). Quantitative PCR analysis using a probe downstream of the gene trap integration site confirmed loss of wild-type transcript in mutant embryos. (J:212179)
