An insertion vector containing a splice acceptor, beta-geo (a fusion of beta-galactosidase and neomycin resistance genes), and a polyadenylation sequence was inserted into the intron between exon 1 and exon 2 of the gene. qPCR analysis confirmed the absence of mRNA expression in the peripheral blood, thymus, spleen, and lymph nodes of homozygous mutant mice.
