A loxP-flanked neomycin cassette was inserted into intron 10 and a TAP-tag cassette was inserted in-frame into exon 11 at the end of the coding sequence. The neomycin cassette was removed after targeting by transient expression of cre recombinase. The TAP epitope tag did not affect expression of function p53 protein produced as determined by numerous biochemical and phenotype analysis on homozygote mice. (J:150816)
