A single loxP site before exon 5 and a PGK-neo cassette flanked by two frt sites followed by a loxP site after exon 5 was introduced into the locus via homologous recombination. The exon 5 genomic region encoding the putative 4th transmembrane domain was deleted in ES cells by transient cre expression. RT-PCR analysis of transcripts from whole brain confirmed the absence of exon 5. Immunoprecipitation and Western blot analyses confirmed loss of protein product. (J:148915)
