A floxed neomycin cassette replaced 4 kb of genomic sequence that included the V5 gene segment and the intervening sequence that lies between the V5 gene segment and the TEA promoter. Cre-mediated deletion of the neomycin cassette removed the V5 gene segment and brought the TEA promoter into the same genomic position as the V5 gene segment. Correct targeting was confirmed by Southern blotting. (J:149413)
