A floxed neomycin cassette was placed upstream of exon 4 with an additional loxP site placed downstream of exon 5. The neomycin cassette was removed by transient expression of cre recombinase leaving exons 4 and 5 floxed. Exon 4 encodes the DSL region, which is conserved across species and is known to be crucial for ligand-receptor interactions. Deletion of exon 4 and exon 5 by cre recombinase was predicted to result in a frameshift mutation and premature truncation of the protein after 192 amino acids. (J:149131)
