A BAC clone containing the Nkx2-5 locus was modified by homologous recombination in E. coli. A targeting vector containing the hrGFP reporter, FRT-flanked kanamycin/TK selection cassette, and Nkx2-5 3' and 5' homology arms was used to replace exon 1 and intron 1 of the Nkx2-5 locus via recombination in red-alpha and red-beta recombinase expressing cells. The selection cassette was excised from recombinant clones by induction of lambda PR-Flp in the bacterial cells. The recombined BAC DNA was microinjected into C57 mice.
