A floxed neo cassette was inserted in intron 1 and an A(-1)T signal peptide mutation was inserted in exon 1 via homologous recombination. The floxed neo cassette was then removed by Cre mediated recombination. RT-PCR confirmed the expression of mutant transcript in the hypothalamus. (J:148012)
