A luciferase cDNA fused in-frame with the noncoding part of exon 2 gene was preceded by a floxed neomycin expression cassette that contained a synthetic polyadenylation sequence to stop transcription. This construct was inserted downstream of exon 2 with an additional loxP site inserted upstream of the exon. Upon cre recombinase mediated excision of exon 2 and the neo cassette, luciferase will be expressed under the control of endogenous locus. (J:147139)
