A farnesylated form of EGFP was inserted in frame with the two first amino acids of the gene locus. Half of the exon 1 and the entire exon 2 were eliminated with transcription following EGFP terminated by a strong polyadenylation signal. A loxP flanked neomycin cassette was included downstream of EGFP for targeting purposes. Gene inactivation was confirmed by a lack of protein expression by homozygote splenic CD8alpha+ dendritic cells. GFP expression was detected in the same population of dendritic cells. (J:147472)
