A floxed neomycin cassette was placed upstream of exon 5 in a Hind-III restriction site, and a loxP site was placed downstream of exon 7 in a Bam-HI site. Correct targeting was confirmed by Southern blotting. Gene expression was unaffected as demonstrated by in situ staining of small intestine epithelial cells. (J:146640)
