The Ple54-EGFP/NLS transgene (pEMS1200) was designed with the 3687 bp Ple54 minipromoter (DCX-C; derived from a portion of the promoter and two downstream putative regulatory elements from the human doublecortin (DCX) gene) upstream of a minimal F5 mutant-frt site, an enhanced green fluorescent protein (with mutated TAA->TTA stop), a nuclear localization signal, a second minimal frt site, an SV40 early polyA signal, and a human HPRT complementary sequence (containing exon1, intron1, exon2, and part of intron2). This construct was targeted as a single copy knockin to the Hprtb-m3 mutant locus on the X chromosome. Detection of EGFP is strongest in soma of cells in the neurogenic regions of the adult brain: the subgranular zone of the dentate gyrus as well as the forebrain subventricular zone (rostral migratory stream and olfactory bulb). Additionally, EGFP-positive cell bodies are detected in the neuropil of the cortex, hippocampus and brainstem. Double label immunocytochemistry shows that the EGFP-positive cells of the forebrain subventricular zone and the rostral migratory stream co-express Doublecortin. (J:144244, J:164356)
