The transgene is a bacterial artificial chromosome (BAC), RP24-330G11, bearing a 186 kb mouse genomic segment that contains the Kit oncogene, including the 32 kb first intron, and ~77 kb of 5' and ~ 99 kb of 3' DNA flanking the gene, into which the coding sequence for enhanced green fluorescent protein (EGFP) has been inserted at the Kit initiation codon; a single Frt site remains associated with the EGFP insert. Neonatal mice exhibit intense skin fluorescence; gross UV examination and immunohistologic analysis for EGFP reveals adult EGFP expression in organs and specific cell types consistent with the known KIT expression pattern. Coexpression of EGFP and of KIT from the endogenous gene have been demonstrated in cell groups, although not always in the same individual cells, of the heart. (J:144922)
