A transcriptional stop signal was inserted upstream of a hygromycin cassette which flanked upstream by lox511 and downstream by a loxP site in reverse orientation. The resulting properly targeted ES cells were then used for RCME (recombinase mediated cassette exchange). The exchange cassette contained a lox522 site followed in order by a U6 PolIII promoter, and shRNA duplex corresponding to a 19-mer in exon 10 of Bmpr2, a neomycin selection cassette, and finally a reverse orientation loxP site. Cre mediated cassette exchange was then performed in ES cells. (J:145514)
