Transgenic mice were created using a 270 kb YAC (CEPH clone 366H2) bearing a genomic DNA segment from human Chromosome 6 containing the lipoprotein, Lp(a)-like 2 gene; a 110 kb variant of the gene encoding lipoprotein, Lp(a); and the plasminogen gene. The LPA allele was determined to have 12 tandem copies of the variable copy number, 5.5 kb kringle IV-like repeat. Transgenic mice were shown by northern blot and RT-PCR analysis to express LPA mRNA in the liver and by immunoblot analysis to have significant (up to 75 mg/dL) plasma LPA; expression is suppressed by normal adult male levels of testosterone.
