A bacterial artificial chromosome containing the Il17f locus was used to construct a transgene vector. Exons 1 and 2 of the locus were replaced with a cre recombinase expression cassette and a neomycin selection cassette. Founder mice were obtained after injection of linearized vector into the pronuclei of fertilized eggs. When transgenic mice were crossed to mice carrying the cre-reporter allele Gt(ROSA)26Sortm1(EYFP)Cos, cre expression was detected in CD8+ T cells and, to a lesser extent, CD4+ T cells after culturing in vitro under Th17-polarizing conditions.
