The transgene comprises 6 kilobases of mouse genomic DNA containing the upstream regulatory elements of the dentin sialophosphoprotein (Dspp) gene, including the noncoding first exon and the first intron, followed by a 1.5 kb cDNA encoding a constitutively active porcine transforming growth factor beta 1, in which Cys223 and Cys225 have been replaced by serine, and an SV40 polyadenylation signal. Transgenic mouse lines with independent transgene integration sites were produced from 5 transgenic founder animals. This record represents the line of mice with a high level of transgene expression. TGFB1 is expressed from this transgene predominantly in the odontoblasts. (J:118918)
