This transgene was created by coinjection of three overlapping fragments. The first fragment (SphI-SphI restriction fragment) contained 15 kb of sequence 5' to the Atoh1 coding sequence. The second (NcoI-NcoI) contained up- and downstream sequence flanking the coding sequence where the coding sequence between the first ~2.5 kb and the last ~1.5 kb was replaced in-frame with the beta-galactosidase reporter gene (without NLS sequence). The third sequence (BglII-BglII) contained the last ~400 bp of coding sequence and 6 kb 3' of the coding region. Five individuals were identified which expressed lacZ in a pattern consistent with the promoter although the expected order of recombination was not confirmed. One of these lines was maintained.
