Using ES cells containing Igf1tm1Arge in which exon 4 was replaced with a neo cassette, exon three was replaced with a full-length cDNA of the gene preceded by a floxed puromycin resistance cassette and three polyadenylation sequences from SV40. The full-length cDNA is only expressed when the polyadenylation sites have been removed. (J:142247)
