Transient cre expression in ES cells was used to remove the floxed exon 3 and the neo cassette. This deletion results in a frame shift and premature termination of translation at amino acid 168 of 840. A two-fold reduction in protein expression levels was confirmed by western blot analysis on embryonic fibroblasts heterozygous for the allele. (J:141633)
