Yellow fluorescent protein attached to the palmitoylation domain of neuromodulin (YFP-Mem) was inserted into the first coding region of the targeted gene and its expression was driven by the Runx3 distal promoter. A PGK-neomycin resistance cassette flanked by loxP sites was placed within the first exon immediately 3' of YFP. Protein expression of the targeted gene was almost undetectable in CD8+ T cells and was much lower in TH1-polarized activated CD4+ T cells. YFP was expressed in MHC class I restricted thymocytes, by resting CD8+ T cells, and in activated CD4+ T cells. (J:141144)
