Three NF-kappa B response elements derived from the 5' LTR of the HIV genome and a minimal c-fos promoter were placed upstream of a EGFP reporter gene. This reporter construct was inserted upstream of genomic sequence containing the promoter and first two exons of the Hprt gene locus that is deleted in BK ES cells. Correct recombination rescued the Hprt defect, ensuring that only a single copy of the NF-kappaB reporter construct integrated into the Hprt locus. GFP positive mononuclear cells are found in both male and female mice. (J:92020)
