A loxP site was inserted upstream of exon 20 and loxP-flanked neomycin cassette was inserted downstream of exon 20. Correct targeting was confirmed by genomic PCR. The floxed exon contains the polyadenylation signals so that upon expression of cre recombinase, the resulting transcipts derived from this locus would be truncated and have a shorter half-life.
