Exon 1, which contains the translation start, was replaced with a floxed exon 1 and an upstream FRT-flanked neomycin resistance cassette. The neomycin resistance cassette was removed by germline expression of the FLP recombinase after targeting. Upon expression of cre recombinase, exon 1 is excised creating a null allele. (J:140109)
