A targeting vector was constructed to replace the region of exon 3 that encodes amino acids 38 through 73 with a neomycin selection cassette. Correct targeting was confirmed by Southern blotting. Cultured splenocytes failed to produce IL-5 or IL-13 when incubated with the ligand to the gene product. (J:139080)
