A 800-kb yeast artificial chromosome (YAC) was constructed from human germline sequence of the immunoglobulin kappa light chain gene cluster. This YAC consisted of 32 V-kappa gene segments, all 5 J segments, the C-kappa constant region, the downstream kappa deleting element and a neomycin resistance cassette. Tw transgenic lines were created with one, J23.1, being propagated for further analysis. Southern blot analysis confirmed that the YAC inserted without any recombination events. Transgenic mice were found to express human kappa light chain immunoglobulin. (J:70620, J:139719)
