A 970-kb yeast artificial chromosome (YAC) was constructed from human germline sequence of the immunoglobulin heavy chain gene cluster. This YAC consisted of 66 V-heavy gene segments, all 6 J segments, all 30 D gene segments, the C-mu, the C-delta and the C-gamma2 constant regions, a downstream murine enhancer element and a neomycin resistance cassette. Seven transgenic lines were created with two, L10 and J9.2, being propagated for further analysis. Southern blot analysis confirmed that the YAC inserted without any recombination events. Transgenic mice were found to express human heavy chain immunoglobulin. (J:70620, J:139719)
