A rat Erbb2 cDNA containing an activating mutation and point mutations in the catalytic domain is expressed under the control of the MMTV LTR with SV40 polyadenylation and splicing signals. The activating mutation, derived from a clone isolated from mouse mammary tumor tissue, consists of a 12 codon in-frame deletion (neu8142 or NDL1) in the region encoding the extracellular region directly adjacent to the transmembrane domain. In addition, sequences encoding the tyrosine kinase domain were altered to mutate residues 1028 (site A), 1144 (site B), 1201 (site C), 1226/1227 (site D), and 1253 (site E) from tyrosine to phenylalanine. Expression of the transgene in mammary tissue was confirmed by RT-PCR. (J:67363)
