An 8.2 kb construct containing all the mouse (A/J) H2-Ea kappa exons (from clone lambda Charon 4) plus 2 kb of DNA 5' to the cap site and 1.4 kb 3' to the poly(A) addition site was microinjected into (C57BL/6 x SJL) embryos. Two lines 23 and 16 were generated with line 16 carrying 12 copies of the transgene in its genome and line 23 having 4 copies. Northern blots and S1 nuclease analysis showed transcripts in the spleen. Line 16 produced more transcripts than line 4, and both lines showed correct initiation sites of transcription. (J:111128)
