A floxed neo cassette was inserted downstream of exon 4 and an additional loxP site was inserted upstream of exon 3. Cre-mediate recombination in ES cells removed both exon 3, exon 4 and the neo cassette. The absence of transcript was confirmed by RT-PCR and enzymatic assays revealed a less than 5% of wild-type activity level. (J:135143)
