A transgenic BAC vector contained a mouse pro-melanin-concentrating hormone (Pmch; MCH) promoterand the N terminus of the MCH gene controlling a human ataxin 3 (ATXN3) cDNA (encoding from amino acid 286 to the C terminus) containing 7 polyglutamine repeats that was introduced to the ATG site of the MCH BAC. The N terminus of the ATXN3 contained a hemagglutinin (HA) epitope and the C terminus contained a myelocytomatosis (MYC) oncogene epitope followed by the murine protamine 1 gene fragment that provided an intron and polyadenylation signal. The construct was injected into FVB/NJ fertilized oocytes. 5 transgenic lines were established. Line 1 was donated to JAX. Transgene expression in Pmch neurons was verified with double immunohistochemistry using MYC and MCH antibodies. (J:104264)
