The variable regions 3 and 4 were replaced with a floxed neo cassette and a variable region 4 and 2 with a 5' loxP site. Transient cre expression in ES cells was used to remove the neo cassette and the inserted variable region 4 and 2 leaving only the endogenous variable region 2. (J:133974)
