Parts of exon 2 and 3 were replaced with an EGFP expression cassette followed by two polyA stop sequences and a loxP-flanked neomycin resistance cassette. Founder mice were mated with cre transgenic mice to remove the neomycin selection cassette. Gene inactivation was confirmed by immunoblot analysis on bone marrow derived mast cells. Immunoblot analysis also confirmed that the adjacent Tpsab1 gene locus contained the intact 129 allele and not the null C57BL/6 allele. (J:133373)
